The goal of the present study is to investigate role intraamygdaloid oxytocin in learning-related mechanisms. Oxytocin a neuropeptide which involved social bonding, trust, emotional responses and various behaviors. By conducting passive avoidance Morris water maze tests on male Wistar rats, memory performance learning was investigated. doses 10 ng 100 were injected into central nucleus amygdala. Our results showed that significantly reduced time required locate platform during test while increasing latency test. <b><i>Introduction:</i></b> Anxiety disorders are prevalent mental conditions characterized by exaggerated anxious arousal and threat reactivity. Animal human studies suggest an anxiolytic potential of the neuropeptide oxytocin (OT), yet, while a clinical application will require chronic administration protocols, previous have exclusively focused on single-dose (acute) intranasal OT effects. <b><i>Objective:</i></b> To facilitate translation mechanism into application, we determined whether effects maintained with repeated (chronic) or influenced dose frequency trait anxiety. <b><i>Methods:</i></b> In pre-registered double-blind randomized placebo-controlled pharmaco-fMRI trial acute (single dose) as well two different frequencies (OT administered daily for 5 days every other day) emotional reactivity were assessed in <i>n</i> = 147 individuals high versus low anxiety (ClinicalTrials.gov ID: NCT03085654). <b><i>Results:</i></b> produced valence, frequency, anxiety-specific effects, such that low-frequency (intermittent) dosage specifically attenuated neural increase amygdala-insula-prefrontal circuits observed placebo-treated subjects response to threatening but not positive stimuli. <b><i>Conclusions:</i></b> The present provides first evidence low-dose has alleviate elevated levels, suggesting treatment disorders.

Abstract The amygdala is a key region in current neurocircuitry models of reactive aggression as it crucially involved detecting social threat and provocation. An increased reactivity to angry faces has been reported aggression-prone individuals the neuropeptide oxytocin (OT) could dampen anger-related number mental disorders. One example antisocial personality disorder (ASPD) which so far only studied limited numbers. To address question whether OT can normalize hyperreactivity emotional faces, we conducted functional magnetic resonance imaging experiment with 20 men 18 women ASPD male female healthy control (HC) participants double-blind, randomized, placebo (PLC)-controlled within-subject design. Participants were exposed an emotion classification task (fearful, angry, happy faces) after receiving intranasal dose (24 IU) synthetic or PLC. We found attenuate right hyperactivity such extent that intensity activity group condition decreased level PLC HC group. There was also trend effects generally larger than men. These findings suggest differentially modulates following threatening provoking cues dependence psychopathology (ASPD vs. HC) sex (male female).
