Liver cancer is one of the most common malignancies, with severe morbidity and mortality. While considerable progress has been made in liver treatment, 5-year overall survival (OS) patients not improved significantly. Reasons include inadequate capability early screening diagnosis, a high incidence recurrence metastasis, degree tumor heterogeneity, an immunosuppressive microenvironment. Therefore, identification validation specific robust biomarkers are major importance for screening, timely accurate prognosis, prevention progression. In this review, we highlight some latest research potential applications biomarkers, describing hotspots prospective directions biomarker discovery.

The current situation of hepatocellular carcinoma (HCC) management is challenging due to its high incidence, mortality, recurrence and metastasis. Recent advances in gene genetic expression regulation have unveiled the significant role non-coding RNA (ncRNA) various cancers. This led formulation competing endogenous (ceRNA) hypothesis, which posits that both coding ncRNA, containing miRNA response elements (MRE), can share same sequence. results a competitive network between ncRNAs, such as lncRNA mRNA, allowing them regulate each other. Extensive research has highlighted crucial ceRNA HCC development, impacting cellular processes including proliferation, metastasis, cell death, angiogenesis, tumor microenvironment, organismal immunity, chemotherapy resistance. Additionally, network, mediated by or circRNA, offers potential early diagnosis prevention HCC. Consequently, ceRNAs are emerging therapeutic targets for complexity these networks aligns with multi-target approach traditional Chinese medicine (TCM), presenting novel perspective TCM combating Research beginning show compounds prescriptions affect progression through inhibiting proliferation inducing apoptosis. Currently, lncRNAs TUG1, NEAT1, CCAT1, along their associated networks, among most promising ncRNAs research. However, this field still infancy, necessitating advanced technology extensive basic fully understand mechanisms treatment.

Hepatocellular carcinoma (HCC) is the most common form of liver cancer, accounting for ~90% neoplasms. It second leading cause cancer-related deaths and seventh cancer worldwide. Although there have been rapid developments in treatment HCC over past decade, incidence mortality rates remain a challenge. With widespread use hepatitis B vaccine antiviral therapy, etiology shifting more toward metabolic-associated steatohepatitis (MASH). Early-stage can be treated with potentially curative strategies such as surgical resection, transplantation, radiofrequency ablation, improving long-term survival. However, patients, when diagnosed, are already intermediate or advanced stages. Molecular targeted followed by immune checkpoint inhibitor immunotherapy, has revolution systemic treatment. Systemic especially patients compromised function still challenge due to significant resistance blockade, tumor heterogeneity, lack oncogenic addiction, effective predictive therapeutic biomarkers.

Abstract Introduction Tumor‐associated macrophages, a major component of the tumor microenvironment, undergo polarization into M2 macrophages (M2), and thereby exert an immunosuppressive effect to induce cancer metastasis. This study strives uncover molecular mechanism underlying this event in hepatocellular carcinoma (HCC). Methods Proteasome subunit alpha 5 (PSMA5) expression liver (LIHC) tissues its association with LIHC patients were predicted using StarBase. PSMA5 level human HCC cells was manipulated via transfection. Exosomes isolated from cells, internalized which cocultured cells. Exosome internalization observed after fluorescence labeling. cell migration invasion evaluated by wound healing Transwell assays. Xenograft assay performed investigate role vitro tumorigenesis. assessed enzyme‐linked immunosorbent assay, quantitative reverse transcription polymerase chain reaction, immunohistochemistry. exosomes Janus Kinase 2 (JAK2)/signal transducer activator 3 (STAT3) activation tumors detected Western blot analysis. Results High associated compromised survival patients. knockdown inhibited invasion. downregulated these cell‐isolated successfully facilitated JAK2/STAT3 pathway activation. cell‐secreted exosomal exosome‐induced on attenuated promotion induced exosome‐treated migration/invasion tumorigenesis along vivo Conclusion hinders suppress progression restraining signaling.

The digital immunoassay is a highly sensitive detection technique based on single-molecule counting and widely used in the ultrasensitive of biomarkers. Herein, we developed fluorescent microsphere-based (FMDIA) by employing microspheres as both carriers for immunoreaction reports imaging. In this approach, target protein sample was captured to form biotin-labeled sandwich immunocomplex, then, containing molecules were adding streptavidin-coated magnetic beads (SA-MBs). By proportion fluorescence-positive beads, concentration can be precisely quantified. As proof concept, α fetoprotein (AFP) human interleukin-6 (IL-6) assess analytical performance proposed FMDIA, limit (LOD) values 21 pg/mL (0.30 pM) 0.19 (7.3 fM) achieved, respectively. results AFP serum samples patients healthy people consistent with reference given hospital. Furthermore, various colors encoding, FMDIA easily realize simultaneous multiple proteins without need introduce modified beads. This multiplex strategy, which reactions are first carried out then capture reporters molecules, provides new idea assays.
