Accumulating data suggest that chronic neuroinflammation-mediated neurodegeneration is a significant contributing factor for progressive neuronal and glial cell death in age-related neurodegenerative pathology. Furthermore, it could be encountered as long-term consequences some viral infections, including post-COVID-19 Parkinsonism-related sequelae. The current systematic review focused on recent question aroused during the pandemic’s successive waves: are there post-SARS-CoV-2 immune-mediated reactions responsible promoting neurodegeneration? Does host’s dysregulated immune counter-offensive contribute to pathogenesis of diseases, emerging Parkinson’s disease, complex interrelation between genetic epigenetic risk factors? A synthetic literature was accomplished based ”Preferred Reporting Items Systematic Principles Reviews Meta-Analyses” (PRISMA) methodology, registration specific online platform: International prospective register reviews—PROSPERO, no. 312183. Abstract Osteoarthritis (OA) is a chronic joint disease characterized by synovitis and cartilage destruction. The severity of OA highly associated with the imbalance between M1 M2 synovial macrophages. In this study, novel strategy designed to modulate macrophage polarization reducing intracellular reactive oxygen species (ROS) levels regulating mitochondrial function. A ROS‐responsive polymer synthesized self‐assemble astaxanthin autophagy activator rapamycin form nanoparticles (NP@Poly RHAPM ). vitro experiments show that NP@Poly significantly reduced ROS levels. Furthermore, restored membrane potential, increased glutathione (GSH) levels, promoted autophagy, hence successfully repolarizing macrophages into phenotype. This repolarization enhanced chondrocyte proliferation vitality while inhibiting apoptosis. vivo utilizing an anterior cruciate ligament transection (ACLT)‐induced mouse model revealed anti‐inflammatory cartilage‐protective effects , effectively mitigating progression. Consequently, findings suggest intra‐articular delivery nanocarrier systems holds significant promise as potential effective therapeutic for treatment.

Abstract Immune‐mediated diseases (IMDs) are chronic conditions that have an immune‐mediated etiology. Clinically, these appear to be unrelated, but pathogenic pathways been shown connect them. While inflammation is a common occurrence in the body, it may either stimulate favorable immune response protect against harmful signals or cause illness by damaging cells and tissues. Nanomedicine has tremendous promise for regulating treating IMIDs. Various nanoparticles coated with nanotherapeutics recently fabricated effective targeted delivery inflammatory RNA interference (RNAi) offers genetic approach, particularly if traditional treatments ineffective IMDs. In cells, several signaling can suppressed using RNAi, which blocks expression of particular messenger RNAs. Using this molecular undesirable effects anti‐inflammatory medications reduced. Still, there many problems short‐interfering RNAs (siRNAs) treat IMDs, including poor localization siRNAs target tissues, unstable gene expression, quick removal from blood.
