B cells play an important role in humoral immunity regulation, but their allo‐HSCT has not been well studied. Hematopoietic stem and progenitor cells (HSPCs) play a pivotal role in blood cell production, maintaining the health homeostasis of individuals. Dysregulation HSPC function can lead to blood-related diseases, including cancer. Despite its importance, our understanding genes pathways underlying development associated pathological mechanisms remains limited. To elucidate these unknown mechanisms, we analyzed databases patients with disorders performed functional gene studies using zebrafish. We employed bioinformatics tools explore three public focusing on myelodysplastic syndrome (MDS) related model studies. This analysis identified significant alterations several genes, especially SMC2 other condensin-related MDS. further investigate Smc2 hematopoiesis, generated smc2 loss-of-function zebrafish mutants CRISPR mutagenesis. Further analyses revealed that depletion induced G2/M cycle arrest HSPCs, leading their maintenance expansion failure. Aging of hematopoietic and immune system (HIS) leads to cellular senescence dysregulation, contributing age-related diseases. Here, we show that Procyanidin C1 (PCC1), a compound with both senolytic senomorphic properties, can counteract aging-related changes in HIS. Using single-cell RNA sequencing validation experiments, found aging induced senescence, inflammation, dysregulation the bone marrow spleen tissues mice. Long-term PCC1 treatment improved key physiological parameters especially grip strength aged Further analysis revealed PCC1's broad geroprotective effects on HIS, including an increase proportion B cells (BCs) stem (HSCs), suppression senescence-associated markers, restoration normal processes. Moreover, reversed aging-induced alterations HSCs through upregulating Nedd4 CD62L-Ca2+ axis expression.
