In mammals, because cardiomyocytes withdraw from cell-cycle activities shortly after birth, the heart cannot repair damage caused by a myocardial injury; thus, understanding how proliferate is among most important topics in cardiovascular sciences. newborn neonatal when left ventricular injury applied hearts earlier than postnatal day 7, actively and regenerate lost myocardium following weeks. The regulators promoting cardiomyocyte proliferation were discovered analyzing transcriptomic data generated models. Most of these support mRNA production machinery, yet requires translation into functional proteins under regulation RNA-binding (RBPs). this work, we performed meta-analysis to study relationship between RBP expression proliferation. To identify RBPs associated with mouse pig proliferation, single-nuclei RNA sequencing (snRNA-seq) regenerating reanalyzed via an Autoencoder focusing on expression. We also analyzed new bulk RNA-seq two human-induced pluripotent stem cell-derived (hiPSC) (hiPSC-CM) cell lines; first line was harvested sixteen days differentiation, cells still proliferated, second one hundred forty ceased cycle activity. Then, mouse, pig, hiPSC-CM compared across species. Twenty-one found be consistently upregulated, six downregulated proliferating hiPSC-derived cardiomyocytes. Among upregulated species, immunofluorescence-based imaging analysis validated significant increase DHX9, PTBP3, HNRNPUL1, DDX6 CMs. This all species demonstrated strong

The post-transcriptional regulation of gene expression plays an important role in heart development and disease. Cardiac-specific alternative splicing, mediated by RNA-binding proteins, orchestrates the isoform switching proteins that are essential for cardiomyocyte organization contraction. Dysfunctions impair cause main types cardiomyopathies, which represent a heterogenous group abnormalities severely affect structure function. In particular, mutations RBM20 RBFOX2 associated with dilated cardiomyopathy, hypertrophic or hypoplastic left syndrome. Functional analyses different animal models also suggest possible roles other cardiomyopathies because their involvement organizing cardiac programming. Recent studies have provided significant insights into causal relationship between cardiovascular diseases. They show potential correcting pathogenic to rescue cardiomyopathy promote regeneration. Therefore, emerged as promising targets therapeutic interventions dysfunction. challenge remains decipher how they coordinately regulate temporal spatial target genes ensure function homeostasis. This review discusses recent advances understanding implications several well-characterized aim identifying research gaps further investigation this field.

RNA binding proteins (RBPs) post-transcriptionally regulate gene expression by associating with regulatory sequences in the untranslated regions of mRNAs. Cold-inducible RBP (CIRP) is a stress-induced that was recently shown to modulate inflammation response cellular stress, where it increases or decreases pro-tumorigenic (proinflammatory) cytokines different contexts. CIRP altered several cancers, including breast cancer, but effects on cancer not known. Here, we investigate if alters growth and inflammatory profile tumors. Transgenic mice overexpressing mammary epithelium were crossed PyMT mouse model both early late tumorigenesis assessed. The knockdown also assessed Py2T cell grafts. Overexpression led decreased model. Conversely, grafts increased tumor growth. Luminex cytokine assays environment. Trastuzumab-induced cardiotoxicity (TIC) is a common and serious disease with abnormal cardiac function. Accumulating evidence has indicated some non-coding RNAs (ncRNAs), acting as competing endogenous (ceRNAs), have been reported to affect the cardiovascular process. However, role of ncRNA-mediated ceRNA regulatory mechanisms in TIC remains unclear. The present research aims systematically investigate alterations expressions all ncRNA using whole-transcriptome RNA sequencing. sequencing revealed total 43 circular (circRNA), 270 long noncoding (lncRNA), 12 microRNA (miRNA), 4131 mRNA were aberrantly expressed mice heart treated trastuzumab.
