Abstract Vitex negundo(Family-Verbenaceae) is a remarkable medicinal plant with numerous therapeutic applications. In this study, phytochemical screening was performed in negundoleaf extract, and extraction optimization by applying response surface methodology (RSM). Methanolic extract microwave-assisted (MAE) were found to be appropriate for obtaining the maximum yield of bioactive compounds. single-factor analysis, solvent concentration, microwave power, time variations analyzed calculate amount TPC, TFC, TAC extract. The quantity fairly obtained from 60%–95% (v/v) methanol 540 -900 W 60–90 s time. Furthermore, we also employed (RSM) optimizing negundo leaves considering three key parameters: temperature, duration. Condition at 95% methanol, 720W, 90s TPC (227.6 mg gallic acid equivalent (GAE)/g sample), TFC (978.2 RE/g (82.13 ascorbic equivalents/g sample). characterization negundowas done FTIR GC–MS identify various active groups types constituent’s presence Subsequently, compounds evaluated their bioactivity through molecular docking interactions target proteins in silico. antibacterial activity optimized studied against two Gram +ve (Bacillus sphaericus, Pectobacterium) Gram-ve (Escherichia coli, Pseudomonas aeruginosa) bacteria, inhibitory effect observed all four tested bacteria. highest shown B. sphaericus (19± 0.087) E. coli (19±0.064) followed Pectobacterium (18±0.068) Pseudomonas(13±0.093). hemolytic leaf (at dose 20µg/ml) shows minimum activity, i.e., 1.33± 0.06 %. It may concluded study that potential safe source natural antioxidants can utilized green therapeutics.
