Circular RNAs (circRNAs) are increasingly recognized as significant regulators in multiple cancers, such hepatocellular carcinoma (HCC), frequently affecting microRNA (miRNA) expression. The diagnostic and prognostic roles of circRNAs, specifically circ_0009910 circ_0027478, conjunction with miR-1236-3p, HCC, have not yet been fully investigated. In this pilot study, we assessed the expression levels circ_0009910, miR-1236-3p plasma samples from 100 patients diagnosed HCC 50 healthy controls through reverse transcriptase–quantitative polymerase chain reaction (RT-qPCR). performance was evaluated using receiver operating characteristic (ROC) curve analysis, correlations clinicopathological features were examined. Circ_0009910 circ_0027478 exhibited upregulation (p < 0.05), whereas demonstrated downregulation 0.05). accuracy (area under [AUC] = 0.90), effectively differentiating showing a correlation tumor size, metastasis, alpha-fetoprotein (AFP) Both positive features, an inverse correlation. Logistic regression validated capabilities these biomarkers. results indicate that AFP three, present promising profile for HCC. Abstract Developing messenger RNA (mRNA) vaccines for COVID-19 renewed and intensified the interest in using mRNA disease prevention treatment. Despite their efficacy, linear molecules are short-lived human body, primarily due to enzymatic degradation at free ends. In contrast, circular (circRNA) exhibits enhanced stability resistance exonuclease degradation. However, this depends highly on purity. Unfortunately, vitro transcription/self-splicing reaction products contain a mixture of RNAs. Yet, practical methods purifying circRNA from solutions containing contaminants lacking. study, we explored feasibility ultrafiltration purify protein-encoding produced by self-splicing precursor (preRNA) during transcription (IVT). We measured sieving coefficients, separation metric, circRNA, RNA, nicked conformers polyethersulfone membranes with molecular weight cutoffs 30 300 kDa, analyzing performance as function permeate flux. also estimated critical fluxes determined suitable operating conditions purification. achieved purity 86% yield above 50%. By comparison, size-exclusion high-performance liquid chromatography (SE-HPLC), leading alterative technology, was 41% 45%. These findings highlight superior method research scale. Its scalability suggests that it could play role enabling large-scale manufacturing circRNA-based therapeutics.
