Introduction Colorectal cancer is a leading cause of related-death worldwide, and resistance to 5-fluorouracil (5FU, key component chemotherapy regimens, major clinical concern. We have previously elucidated the effects Rhus coriaria ethanolic extract (RCE) in triple-negative breast cancer, CRC, pancreatic cells. Here, we explored anticancer RCE parental (HCT-116-WT) 5FU-resistant HCT-116 (HCT-116-5FU-R) CRC Methods MTT assay was used assess cell viability. Muse analyzer viability, cycle distribution, apoptosis. Additionally, colony formation growth assays western blots were performed. In vivo assessed by an ovo chick embryo tumor assay. Results found that inhibited viability capacities HCT-116-WT HCT-116-5FU-R The antiproliferative attributed DNA damage-mediated impairment at S phase, induction Beclin-1-independent autophagy both lines. Mechanistically, inhibition mTOR, STAT3 p38 MAPK pathways implicated latter. induced caspase-7-independent apoptosis However, cells resistant through upregulation survivin, downregulation Bax. Using proteasome inhibitors, clarified pathway contributed RCE-mediated death Lastly, confirmed xenografts model. Discussion Collectively, our findings highlight source phytochemicals can be as agents for CRC.

Overcoming the limited bioavailability and extensive metabolism of effective in vitro drugs remains a challenge that limits translation promising into clinical trials. Resveratrol, despite its well-reported therapeutic benefits, is not metabolically stable thus has been utilized as an drug. This because it needs to be consumed large amounts overcome burdens conversion less metabolites. Herein, we summarize more relevant approaches modify resveratrol, aiming increase biological efficacy. We discuss combination therapies, derivatization, use resveratrol nanoparticles. Interestingly, with established chemotherapeutic shown effects on colon cancer (with oxaliplatin), liver cisplatin, 5-FU), gastric doxorubicin). On other hand, derivatizing including hydroxylation, amination, amidation, imidation, methoxylation, prenylation, halogenation, glycosylation, oligomerization, differentially modifies could used for preferential outcomes. Moreover, encapsulation allows trapping within different forms shells targeted therapy. Depending nanoparticle used, can enhance solubility absorption, increasing These include polymers, metals, solid lipids, nanoparticles have preclinical results, adding “hype” research resveratrol. review provides platform compare allow directed better treatment options

Cancer is the world's second most common cause of death, with colon cancer (CC) ranking third in prevalence and cancer-related fatalities. CC originates or rectum influenced by genetic mutations, lifestyle, diet. Although chemotherapy, especially 5-fluorouracil (5-FU), fundamental to treatment has improved patient survival, its benefits are often limited significant side effects emerging drug resistance. Recently, plant-based foods have drawn interest research for their natural bioactive compounds relatively low toxicity. Certain plant not only exhibit anti-cancer activity but may also boost effectiveness 5-FU while reducing adverse effects. Despite evidence suggesting a synergistic effect between 5-FU, this area remains fragmented. This review consolidates current on including fruits, vegetables, legumes, tea bioactives treatment. Besides, it explores mechanisms through which these enhance efficacy reduce effects, evaluates clinical application prospects, identifies challenges future directions. comprehensive analysis aims provide scientific foundation integrated CC, highlighting potential enhancing chemotherapy associated toxicities.

Drug resistance is a major problem in cancer treatment, as it limits the effectiveness of pharmacological agents and can lead to disease progression. Cold atmospheric plasma (CAP) technology that uses ionized gas (plasma) generate reactive oxygen nitrogen species (RONS) kill cells. CAP novel approach for overcoming drug cancer. In recent years, there has been growing interest using enhance chemotherapy drugs. this review, we discuss mechanisms behind phenomenon explore its potential applications treatment. Going through existing literature on cancer, highlight challenges opportunities further research field. Our review suggests could be promising option warrants investigation.

A large body of evidence indicates the existence a complex pathophysiological relationship between cardiovascular diseases and cancer. Mitochondria are crucial organelles whose optimal activity is determined by quality control systems, which regulate critical cellular events, ranging from intermediary metabolism calcium signaling to mitochondrial dynamics, cell death mitophagy. Emerging data indicate that impaired drives myocardial dysfunction occurring in several heart diseases, including cardiac hypertrophy, infarction, ischaemia/reperfusion damage metabolic cardiomyopathies. On other hand, diverse human cancers also dysregulate promote their initiation progression, suggesting modulating homeostasis may represent promising therapeutic strategy both cardiology oncology. In this review, first we briefly introduce physiological mechanisms underlying system, then summarize current understanding about impact dysregulated functions We discuss key increased risk complications secondary main anticancer strategies, highlighting potential strategies aimed at alleviating impairment-related tumorigenesis. It hoped summary can provide novel insights into precision medicine approaches reduce cancer morbidities mortalities.

The emergence of chemoresistant disease during chemotherapy with 5-Fluorouracil-based (5-FU-based) regimens is an important factor in the mortality metastatic CRC (mCRC). causes 5-FU resistance are multi-factorial, and besides DNA mismatch repair deficiency (MMR-D), there no widely accepted criteria for determining which patients not likely to be responsive 5-FU-based therapy. Thus, a need systematically understand mechanistic basis treatment failure urgent develop new approaches circumventing major resistance. In this manuscript, we review mechanisms emphasis on: (1) altered anabolic metabolism limiting formation primary active metabolite Fluorodeoxyuridylate (5-Fluoro-2'-deoxyuridine-5'-O-monophosphate; FdUMP); (2) elevated expression or activity enzymatic target thymidylate synthase (TS); (3) dysregulated programmed cell death as Importantly, these can potentially overcome through use next-generation fluoropyrimidine (FP) polymers (e.g., CF10) that display reduced dependence on more potent TS inhibitory activity.
