Immune responses induced by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection play a critical role in the pathogenesis and outcome of disease 2019 (COVID-19). However, dynamic profile immune postinfection SARS-CoV-2 variants concern (VOC) is not fully understood. In this study, peripheral blood mononuclear cells single-cell sequencing was performed to determine profiles response Prototype, Alpha, Beta, Delta rhesus monkey model. Overall, all strains dramatic changes both cellular subpopulations gene expression levels at 1 day (dpi), which associated function including adaptive response, innate immunity, IFN response. COVID-19-related genes revealed different dpi among four strains, reported COVID-19 patients with increased risk autoimmune rheumatic diseases. Delta-infected animal showed inhibition translation pathway. B cells, T monocytes much commonality rather than specificity strains. Monocytes were major responders infection, lasted longer Alpha other Thus, study reveals early Proto or its nonhuman primates, important information for controlling rapidly evolving viruses.

Abstract Cellular mechanotransduction, a critical regulator of numerous biological processes, is the conversion from mechanical signals to biochemical regarding cell activities and metabolism. Typical cues in organisms include hydrostatic pressure, fluid shear stress, tensile force, extracellular matrix stiffness or tissue elasticity, viscosity. Mechanotransduction has been expected trigger multiple such as embryonic development, repair regeneration. However, prolonged excessive stimulation can result pathological multi-organ fibrosis, tumorigenesis, cancer immunotherapy resistance. Although associations between normal homeostasis diseases have identified, regulatory mechanisms among different are not yet comprehensively illustrated, no effective therapies currently available targeting cue-related signaling. This review systematically summarizes characteristics typical conditions with updated evidence. The key effectors responding stimulations listed, Piezo channels, integrins, Yes-associated protein (YAP) /transcriptional coactivator PDZ-binding motif (TAZ), transient receptor potential vanilloid 4 (TRPV4). We also reviewed signaling pathways, therapeutic targets cutting-edge clinical applications related cues.

•Insights into the intricate facets of immune microenvironment hold key to pioneering clinical strategies in combatting bacterial infections.•The design principles for antimicrobial biomaterials vary depending on at different stages infection.•Immunomodulatory display robust efficacy and vaccine attributes animals trials, promising intractable infections. Bacterial infectious diseases are one leading causes death worldwide. Even with use multiple antibiotic treatment strategies, 4.95 million people died from drug-resistant infections 2019. By 2050, number deaths will reach 10 annually. The increasing mortality may be partly due heterogeneity infection microenvironment, such as bacteria, biofilms, persister cells, intracellular small colony variants. In addition, complexity makes direct activity unsatisfactory long-term chronic attributed failing modulate active action cells. Therefore, there is an urgent need effective alternatives treat Accordingly, development immunomodulatory has recently received considerable interest; however, a comprehensive review their research progress lacking. this review, we focus mainly future perspectives used infection. First, describe characteristics acute phases Then, highlight corresponding advantages disadvantages. Moreover, discuss biomaterial-mediated vaccines' potential applications challenges activating innate adaptive memory. This serve reference studies develop next-generation accelerate translation practice.
