The nervous system governs both ontogeny and oncology. Regulating organogenesis during development, maintaining homeostasis, promoting plasticity throughout life, the plays parallel roles in regulation of cancers. Foundational discoveries have elucidated direct paracrine electrochemical communication between neurons cancer cells, as well indirect interactions through neural effects on immune stromal cells tumor microenvironment a wide range malignancies. Nervous system-cancer can regulate oncogenesis, growth, invasion metastatic spread, treatment resistance, stimulation tumor-promoting inflammation, impairment anti-cancer immunity. Progress neuroscience may create an important new pillar therapy.

Some patients remain unwell for months after "recovering" from acute COVID-19. They develop persistent fatigue, cognitive problems, headaches, disrupted sleep, myalgias and arthralgias, post-exertional malaise, orthostatic intolerance other symptoms that greatly interfere with their ability to function can leave some people housebound disabled. The illness (Long COVID) is similar myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) as well persisting illnesses follow a wide variety of infectious agents following major traumatic injury. Together, these are projected cost the U.S. trillions dollars. In this review, we first compare ME/CFS Long COVID, noting considerable similarities few differences. We then in extensive detail underlying pathophysiology two conditions, focusing on abnormalities central autonomic nervous system, lungs, heart, vasculature, immune gut microbiome, energy metabolism redox balance. This comparison highlights how strong evidence each abnormality, illness, helps set priorities future investigation. review provides current road map literature biology both illnesses.

Contemporary studies have completely changed the view of brain immunity from envisioning as isolated and inaccessible to peripheral immune cells an organ in close physical functional communication with system for its maintenance, function, repair. Circulating reside special niches brain's borders, choroid plexus, meninges, perivascular spaces, which they patrol sense a remote manner. These niches, together meningeal lymphatic skull microchannels, provide multiple routes interaction between system, addition blood vasculature. In this Review, we describe current ideas about their implications aging, diseases, immune-based therapeutic approaches.

Abstract In addition to its canonical function of protection from pathogens, the immune system can also alter behaviour 1,2 . The scope and mechanisms behavioural modifications by are not yet well understood. Here, using mouse models food allergy, we show that allergic sensitization drives antigen-specific avoidance behaviour. Allergen ingestion activates brain areas involved in response aversive stimuli, including nucleus tractus solitarius, parabrachial central amygdala. requires immunoglobulin E (IgE) antibodies mast cells but precedes development gut inflammation. ability allergen-specific IgE promote cysteinyl leukotrienes growth differentiation factor 15. Finally, a comparison C57BL/6 BALB/c strains revealed strong effect genetic background on These findings thus point probably evolved niche selection avoid unfavourable environments.
