Vitexin (VI) is a naturally occurring flavonoid derived from the leaves and seeds of Vitex, recognized for its strong antioxidant properties. This study aims to explore effects on renal ischemia-reperfusion injury (IRI) investigate underlying mechanisms. We utilized hypoxia-reoxygenation (H/R) models with HK-2 cell lines (I/R) in mice, applying vitexin preconditioning assess influence IRI. Our findings reveal that mitigated oxidative stress, decreased apoptosis, reduced expression damage indicators such as kidney molecule-1 (KIM-1) neutrophil gelatinase-associated lipocalin (NGAL), along an overall improvement function. To further mechanism, we used network pharmacology molecular docking techniques predict potential targets Results Western blotting immunofluorescence assays indicate may promote mitophagy by suppressing phosphorylation pivotal p38 protein p38/MAPK signaling pathway, offering protection against The could potentially be therapeutic agent alleviate

ABSTRACT Diabetic kidney disease (DKD) is a major diabetic microvascular complication that still lacks effective therapeutic drugs. Ferroptosis recently identified form of programmed cell death triggered by iron overload. It characterized unrestricted lipid peroxidation and subsequent membrane damage found in various diseases. Accumulating evidence has highlighted the crucial roles overload ferroptosis DKD. Here, we review metabolism biology ferroptosis. The role aberrant inducing diverse renal intrinsic death, oxidative stress, fibrosis DKD summarized, elaborate on critical regulatory factors related to Finally, focused significance treatment highlight recent data regarding novel activities some drugs as inhibitors DKD, aiming provide new research targets strategies

Flavonoids, abundant in the human diet, have been extensively studied for their therapeutic bioactivities. Recent research has made significantly advances our understanding of biological activities flavonoids, demonstrating effects various chronic diseases. However, generally low bioavailability flavonoids limits effectiveness. Therefore, it is essential to explore pharmacokinetics paying particular attention roles transporters and metabolizing enzymes. This paper reviews recent studies on bioactivity highlighting importance metabolic enzymes pharmacokinetics.

Ferroptosis is a regulated, iron-dependent form of cell death driven by lipid peroxidation and distinct from apoptosis, necroptosis, pyroptosis. Recent studies implicate ferroptosis as central contributor to the pathogenesis renal fibrosis, hallmark chronic kidney disease associated with high morbidity progression end-stage failure. This review synthesizes current evidence linking ferroptotic signaling fibrotic remodeling in kidney, focusing on iron metabolism dysregulation, glutathione peroxidase 4 (GPX4) inactivation, peroxide accumulation, ferroptosis-regulatory pathways such FSP1-CoQ10-NAD(P)H GCH1-BH4. We detail how tubular epithelial cells modulates pro-fibrotic cytokine release, macrophage recruitment, TGF-β1-driven extracellular matrix deposition. Moreover, we explore therapeutic vulnerability highlighting promising agents including chelators, GPX4 activators, anti-lipid peroxidants, exosome-based gene delivery systems. By consolidating emerging preclinical data, this provides comprehensive mechanistic framework identifies translational opportunities for targeting disease.
