This study optimized the extraction process of crude polysaccharides from Panax notoginseng leaves (PNLP) using ultrasonic-assisted dual-enzyme method through a single-factor combined with response surface experiment. The were subsequently purified and isolated DEAE-Cellulose 52, followed by structural analysis, evaluation antioxidant activity, examination digestive enzyme inhibition. hypoglycemic effects components further clarified. results indicated that polysaccharide had an yield 17.13 ± 0.29%. fraction PNLP-3 (eluted 0.3 M NaCl) was obtained 52 chromatography, exhibiting total sugar content 81.2% molecular weight 16.57 kDa. PNLP is primarily composed arabinose, galactose, galacturonic acid, molar percentages 20.24%, 33.54%, 24.27%, respectively. mainly arabinose 29.97% 49.35%, In this IC50 values for α-glucosidase α-amylase inhibition 1.045 mg/mL 9.53 mg/mL, Molecular docking confirmed exhibits better inhibitory activity against α-glucosidase. Furthermore, alleviated hyperglycemia in insulin-resistant HepG2 cells enhancing glucose consumption glycogen synthesis. exhibited positive correlation its concentration, potentially contributing to reducing oxidative stress. These findings underscore therapeutic potential leaf managing type 2 diabetes offer new perspectives on use natural regulating blood glucose.

In this research, the extraction process of polysaccharides from Pithecellobium clypearia Benth (PCBPs) was optimized using dual-frequency ultrasound-assisted (DUAE). The biological activities PCBPs were investigated by in vitro antioxidant, hypoglycemic, and anti-hyperlipidemic assay. High-performance anion-exchange chromatography, high-performance gel permeation SEM, UV–Vis spectroscopy, FT-IR spectra used to analyze monosaccharide composition, molecular weight, microscopic morphology, characteristic structure PCBPs. results showed that maximum rate 9.90 ± 0.16 % when ultrasonic time 8 min, liquid-to-material ratio 32 mL/g, power 510 W. also possessed excellent activities. addition, average weight 15.07 kDa. consisted rhamnose, arabinose, galactose, glucose, xylose, mannose, glucuronic acid, with molar ratios 11.07 %, 18.54 48.17 10.44 4.62 4.96 2.20 respectively. Moreover, SEM mainly a fine spherical mesh structure. above studies provided valuable theoretical basis for subsequent in-depth study
