Methotrexate (MTX), a primary treatment for moderate to severe psoriasis, is limited in clinical use due suboptimal results and side effects from subcutaneous (SC) injection oral administration. Microneedles offer promising alternative direct MTX delivery targeted skin lesions, but issues such as drug wastage, dosage inaccuracy, residence time the lesions remain. This study introduces tip-swellable microneedle array patch (TSMAP) using photo-cross-linked methacrylated hyaluronic acid (MeHA) biocompatible resin effective loading sustained delivery. A two-cast micromolding with vacuum drying employed concentrate cross-linked MeHA about 30% of needle's height at tip, thereby ensuring that only TSMAP tip swells. Efficient into tips achieved through 30 s solution immersion 10 min drying, potentially minimizing waste incomplete insertion elasticity. Psoriasis, affecting 2–3% of the global population, is a chronic inflammatory skin condition without definitive cure. Current treatments focus on managing symptoms. Recognizing need for innovative drug delivery methods to enhance patient adherence, this study explores new approach using calcipotriol monohydrate (CPM), primary topical treatment psoriasis. Despite its effectiveness, CPM's therapeutic potential often limited by factors like greasiness applications, poor permeability, low retention, and lack controlled delivery. To overcome these challenges, introduces CPM in form nanosuspensions (NSs), characterized an average particle size 211 ± 2 nm. These NSs are then incorporated into trilayer dissolving microneedle patch (MAP) made from poly(vinylpyrrolidone) w poly(vinyl alcohol) as needle arrays prefrom 3D printed polylactic acid backing layer.
