The effectiveness of pyrethroid insecticides is seriously threatened by knockdown resistance (kdr), which induced in insects inherited single-nucleotide polymorphisms the voltage-gated sodium channel (VGSC) gene. VGSC's L1014F substitution results classic kdr mutation, found many pest species. Other substitutions L1014 locus, such as L1014S, L1014C, L1014W, and L1014H, were also reported. In 2022, a new amino acid substitute L1014S Blattella germanica was first discovered China. We modified BgNav1-1 from cockroaches with mutation to study how sensitivity gating affected Xenopus oocytes. reduced half-maximal activation voltage (V1/2,act) -19.0 (wild type) -15.5 mV while maintaining dependency activation. Moreover, dependence inactivation hyperpolarizing shifts -48.3 -50.9 mV. However, compared wild type, did not cause significant shift half (V1/2,act). Notably, unaffected greatly mutation. Tail currents are two types pyrethroids (1 μM): type I (permethrin, bifenthrin) II (deltamethrin, λ-cyhalothrin). All four produced tail currents, differences percentages modifications between variants types. Further computer modeling showed that allosterically modifies binding action on B. VGSC, some residues playing critical role binding. This elucidated mechanism predicted may confer risk resistance, providing molecular basis for understanding mechanisms conferred mutations at 1014 site VGSC.

The encapsulation of pesticides within nanoparticles is a promising approach advanced technology in sustainable agriculture. Lambda-cyhalothrin (LC) was encapsulated by the ionotropic gelation technique into chitosan (CS)/tripolyphosphate (TPP) and CS/alginate (ALG) matrixes. CS-LC were characterized, their efficacy then evaluated against key pest soft fruits Europe United States, Drosophila suzukii. efficiency (74%), nanoparticle yield (80%), polydispersity index (0.341), zeta potential (-23.1 mV) particle size (278 nm) determined at optimum conditions. FTIR confirmed cross-linkage between CS TPP/ALG nanoparticles. formula recommended fractional factorial design associated with formulation variables high or low molecular weight, cross-linking agent (TPP), LC concentration (1.5% w/v) stirring rate (1500 rpm), showing highest desirability value (0.5511). lowest exhibited percent mortality D. suzukii males (86%) females (84%), exceeding that caused commercial product (Karate-zeon® 10% CS) 2 HAT. This first work to use ionic make nanoparticles, best our knowledge. chemical biodegradable polymeric could be helpful for establishing IPM strategy benefits human environmental health lifetime pesticides.
