This study aimed to identify the cytokine expression profile in prostate cancer (PCa) patients compared healthy individuals. Plasma samples from 75 PCa and 14 controls were analyzed using Multiplex ELISA (Luminex) measure levels of 12 cytokines: IL-4, IL-5, IL-6, IL-10, IL-1β, IL-17A, IL-12p70, MCP-1/CCL2, MIP-1α/CCL3, MIP-1β/CCL4, TNF-α, IFN-γ. Differences Mann–Whitney test, Wilcoxon’s rank-sum Spearman’s rank correlation, K-means Clustering unsupervised machine learning validate patterns. In patients, IFN-γ, interleukins exhibited significantly higher levels; conversely, TNF-α MCP-1/CCL2 both had decreased The clustering analysis confirmed that exclusively exhibit highest associations with IFN- γ, IL-5. Furthermore, specific correlations between IL-4 MIP-1 beta, IFN-gamma, IL-5 IFN-gamma did not occur Such results will guide forthcoming vitro vivo human cancer-drug treatment models, paving way for exploration future drug targets candidates potential predict FDA-approved responses by targeting oncogenesis pathways.

Abstract Given that hypoxia is a persistent physiological feature of many different solid tumors and key driver for cancer malignancy, it thought to be major target in treatment recently. Tumor-associated macrophages (TAMs) are the most abundant immune cells tumor microenvironment (TME), which have large impact on development immunotherapy. TAMs massively accumulate within hypoxic regions. represent deadly combination because has been suggested induce pro-tumorigenic macrophage phenotype. Hypoxia not only directly affects polarization, but also an indirect effect by altering communication between macrophages. For example, can influence expression chemokines exosomes, both profound impacts recipient cells. Recently, demonstrated intricate interaction TME relevant poor prognosis increased malignancy. However, there no comprehensive literature reviews molecular mechanisms underlying hypoxia-mediated TAMs. Therefore, this review aim collect all recently available data topic provide insights developing novel therapeutic strategies reducing effects hypoxia.

Abstract KRAS is one of the most mutated genes, driving alternations in metabolic pathways that include enhanced nutrient uptaking, increased glycolysis, elevated glutaminolysis, and heightened synthesis fatty acids nucleotides. However, beyond mechanisms KRAS-modulated cancer metabolisms remain incompletely understood. In this review, we aim to summarize current knowledge on KRAS-related alterations cells explore prevalence significance mutation shaping tumor microenvironment influencing epigenetic modification via various molecular activities. Given rely these changes sustain cell growth survival, targeting processes may represent a promising therapeutic strategy for KRAS-driven cancers.
