ABSTRACT Background Metabolic Dysfunction‐Associated Steatotic Liver Disease (MASLD) encompasses a spectrum of histological conditions ranging from simple steatosis to fibrosing steatohepatitis, and is risk factor for cardiovascular diseases (CVD). While oxidised apolipoproteins A B have been linked obesity CVD, the association between other MASLD yet be established. To fill this gap, we characterised circulating serum peptidome patients with MASLD. Methods We studied 87 biopsy‐confirmed 20 age‐ sex‐matched control (CTRL) subjects. first employed an untargeted LC‐MS/MS peptidomics approach (9 CTRL, 32 MASLD) identify key hits differentially modulated, subsequently validated most relevant findings through targeted in enlarged study population (87 CTRL). Results Untargeted identified several apolipoprotein peptide fragments, including ApoE ApoC‐III, significantly upregulated compared CTRL. Specifically focusing on oxidative status intact its major glycoforms (ApoC‐III 0 , ApoC‐III i ii ), observed marked reduction non‐oxidised forms these peptides alongside substantially increased levels their proteoforms versus controls (but not within disease stages). Oxidised fragments were also correlated obesity, insulin resistance, dyslipidaemia transaminases, suggesting potential link oxidation systemic/hepatic metabolic dysfunction. Conclusion Our data reveals previously unreported profile associated The functional clinical implications warrant further mechanistic investigation.

GLP-1 based therapy represents a new treatment option for inflammatory bowel disease. Ban-Lan-Gen (BLG) granule, known anti-viral TCM formulation, exhibits potential anti-inflammatory activities in treating various kinds of inflammation. However, its effect on colitis and the underlying mechanisms remain unknown.Dextran sulfate sodium (DSS)-induced chronic relapsing mice was established. The disease activity index, histological sign damage, levels proinflammatory cytokines were performed to assess protective effects BLG. Serum level colonic Gcg, GPR41 GRP43 expression, community compositions gut microbiota, SCFAs feces release from primary murine colon epithelial cells characterize BLG microbiota SCFA derived-GLP-1 production.BLG significantly alleviated body weight loss, DAI, shortening, tissue pro-inflammatory cytokine TNF-α, IL-1β IL-6 tissues. Moreover, could observably restore expression serum colitic mice, as well correct alteration by increasing abundances SCFA-producing bacteria, eg, Akkermansia Prevotellaceae_UCG-001, decreasing Eubacterium_xylanophilum_group, Ruminococcaceae_UCG-014, Intestinimonas, Oscillibacter. Furthermore, markedly increase mice. In parallel, ex vivo assay also showed that extract BLG-treatment greatly stimulate secretion cells. These findings suggest anti-colitis are achieved at least partly regulating restoring production, has be developed promising agent colitis.
