Significance Sensors of the innate immune system trigger induction inflammatory responses upon infection and cellular stress. One such sensor is inflammasome pathway, which best described for its role in production cytokines IL-1β IL-18. Other effector functions pathway remain less well defined, here we show that activation this leads to mitochondrial damage dismantling organelle. We link another inflammasome-regulated process called pyroptosis, a proinflammatory form cell death. In summary, characterize during relevance host defense other physiological pathophysiological settings.

Summary Cell death is a fundamental biological phenomenon that essential for the survival and development of an organism. Emerging evidence also indicates cell contributes to immune defense against infectious diseases. Pyroptosis form inflammatory programmed pathway activated by human mouse caspase‐1, caspase‐4 caspase‐5, or caspase‐11. These caspases are used host control bacterial, viral, fungal, protozoan pathogens. requires cleavage activation pore‐forming effector protein gasdermin D caspases. Physical rupture causes release pro‐inflammatory cytokines IL ‐1β ‐18, alarmins endogenous danger‐associated molecular patterns, signifying potential pyroptosis. Here, we describe central role pyroptosis in mediating immunity infection clearance

Summary Inflammatory caspases play a central role in innate immunity by responding to cytosolic signals and initiating twofold response. First, caspase‐1 induces the activation secretion of two prominent pro‐inflammatory cytokines, interleukin‐1β ( IL ‐1β) ‐18. Second, either or caspase‐11 can trigger form lytic, programmed cell death called pyroptosis. Pyroptosis operates remove replication niche intracellular pathogens, making them susceptible phagocytosis killing secondary phagocyte. However, aberrant, systemic pyroptosis vivo may contribute sepsis. Emphasizing efficiency inflammasome detection microbial infections, many pathogens have evolved avoid subvert This review focuses on molecular morphological characteristics individual inflammasomes their contribution defense against infection mice humans.

Abstract Programmed cell death (PCD) refers to the way in which cells die depending on specific genes encoding signals or activities. Apoptosis, autophagy, and pyroptosis are all mechanisms of PCD. Among these mechanisms, is mediated by gasdermin family, accompanied inflammatory immune responses. The relationship between cancer complex, effects vary different tissues genetic backgrounds. On one hand, can inhibit occurrence development tumors; other as a type proinflammatory death, form suitable microenvironment for tumor growth thus promote growth. In addition, induction also considered potential treatment strategy. Studies have shown that DFNA5 (nonsyndromic hearing impairment protein 5)/GSDME (Gasdermin-E) mRNA methylation results lower expression levels DFNA5/GSDME most than normal cells, making it difficult activate cells. During malignant tumors, appropriate chemotherapeutic drugs be selected according DFNA5/GSDME, upregulated thereby increasing sensitivity reducing drug resistance. Therefore, induced may play predominant role cancer. Here, we review latest research anti- protumor its applications treatment.
