Antibiotic resistance is an urgent threat to global health. Antidepressants are consumed in large quantities, with a similar pharmaceutical market share (4.8%) antibiotics (5%). While acknowledged as the major driver of increasing antibiotic resistance, little attention paid contribution antidepressants this process. Here, we demonstrate that at clinically relevant concentrations induce multiple antibiotics, even following short periods exposure. persistence was also enhanced. Phenotypic and genotypic analyses revealed enhanced production reactive oxygen species exposure directly associated increased resistance. An stress signature response stimulation efflux pump expression were persistence. Mathematical modeling predicted would accelerate emergence antibiotic-resistant bacteria, persister cells help maintain Overall, our findings highlight risk caused by antidepressants.

Abstract Antimicrobial resistance is a major threat for public health. Plasmids play critical role in the spread of antimicrobial via horizontal gene transfer between bacterial species. However, it remains unclear how plasmids originally recruit and assemble various antibiotic genes (ARGs). Here, we track ARG recruitment assembly clinically relevant by combining systematic analysis 2420 complete plasmid genomes experimental validation. Results showed that across prevalent, 87% ARGs were observed to potentially among 8229 plasmid-borne ARGs. Interestingly, occur mostly compatible within same cell, with over 88% transfers occurring plasmids. Integron insertion sequences drive ongoing acquisition plasmids, especially which IS26 facilitates 63.1% events In vitro experiment validated important involved transferring gentamicin aacC1 Network four beta-lactam (blaTEM-1, blaNDM-4, blaKPC-2, blaSHV-1) shuffling 1029 45 clinical pathogens, suggesting alarming transferred accelerate propagation pathogens. are also able transmit environmental boundaries, terms high-sequence similarities This study demonstrated inter-plasmid universal mechanism ARGs, thus advancing our understanding emergence multidrug-resistant

Phthalate esters (PAEs) are commonly released from plastic pipes in some water distribution systems. Here, we show that exposure to a low concentration (1-10 μg/L) of three PAEs (dimethyl phthalate (DMP), di-n-hexyl (DnHP), and di-(2-ethylhexyl) (DEHP)) promotes Pseudomonas biofilm formation resistance free chlorine. At PAE concentrations ranging 1 5 μg/L, genes coding for quorum sensing, extracellular polymeric substances excretion, oxidative stress were upregulated by 2.7- 16.8-fold, 2.1- 18.9-fold, 1.6- 9.9-fold, respectively. Accordingly, more matrix was produced the polysaccharide eDNA contents increased 30.3-82.3 10.3-39.3%, respectively, relative unexposed controls. Confocal laser scanning microscopy showed stimulated densification (volumetric fraction 27.1 38.0-50.6%), which would hinder disinfectant diffusion. Biofilm verified atomic force microscopy, measured an increase elastic modulus 2.0- 3.2-fold. also antioxidative system, with cell-normalized superoxide dismutase, catalase, glutathione activities increasing 1.8- 3.0-fold, 1.0- 2.0-fold, 1.2- 1.6-fold, This likely protected cells against damage Overall, demonstrate environmentally relevant levels can upregulate molecular processes physiologic changes promote system expression, enhance disinfectants.
