Abstract Circadian disruptions may result in sleep problems, oxidative stress and an altered inflammatory response. These symptoms contribute to PD pathogenesis, despite a lack of direct experimental evidence supporting this relationship. Clock genes are essential drive maintain circadian rhythm. To elucidate the possible role PD, we investigated 132 tag variants eight clock genes. We genotyped these tags within 1,394 Chinese cases 1,342 controls using Illumina GoldenGate chips. discovered that SNPs ARNTL (rs900147, P = 3.33 × 10 −5 , OR 0.80) PER1 (rs2253820, 5.30 −6 1.31) significantly associated with risk. Moreover, positive association rs900147 variant was more robust tremor dominant (TD) ( 3.44 −4 ) than postural instability gait difficulty (PIGD) 6.06 −2 ). The rs2253820 PIGD 5.42 TD 4.2 Haplotype analysis also showed were PD. Imputation identified some which affect gene expression through altering transcription factor binding site. In summary, our findings suggest genetic polymorphisms genes, as well disruptions, pathogenesis.

Abstract Redox biology is at the core of life sciences, accompanied by close correlation redox processes with biological activities. homeostasis a prerequisite for human health, in which physiological levels nonradical reactive oxygen species (ROS) function as primary second messengers to modulate signaling orchestrating multiple sensors. However, excessive ROS accumulation, termed oxidative stress (OS), leads biomolecule damage and subsequent occurrence various diseases such type 2 diabetes, atherosclerosis, cancer. Herein, starting evolution biology, we reveal roles multifaceted modulators mediate sustain homeostasis. In addition, also emphasize detailed OS mechanisms involved initiation development several important diseases. double‐edged sword disease progression suggest two different therapeutic strategies treat redox‐relevant diseases, targeting sources redox‐related effectors manipulate will largely promote precision medicine. Therefore, comprehensive understanding networks under pathological conditions facilitate medicine benefit patients

The human skin is not only a target for the protective actions of melatonin, but also site melatonin synthesis and metabolism, suggesting an important role local melatoninergic system in protection against ultraviolet radiation (UVR) induced damages. While exerts many effects on cell physiology tissue homeostasis via membrane bound receptors, strong UVR-induced damage including DNA repair/protection seen at its high (pharmocological) concentrations indicate that these are mainly mediated through receptor-independent mechanisms or perhaps activation putative nuclear receptors. destructive UVR significantly counteracted modulated by context complex intracutaneous anti-oxidative with UVR-enhanced UVR-independent metabolites. Therefore, endogenous production, together topically-applied exogenous metabolites would be expected to represent one most potent defense systems UV-induced skin. Despite varied etiologies and symptoms, several neurodegenerative diseases-specifically, Alzheimer's (AD), Parkinson's (PD), Huntington's diseases (HDs)-share the common feature of abnormal circadian rhythms, such as those in behavior (e.g., disrupted sleep/wake cycles), physiological processes diminished hormone release) biochemical activities antioxidant production). Circadian disturbances are among earliest symptoms these diseases, molecular mechanisms system suspected to play a pivotal, possibly causal, role their natural histories. Here, we review abnormalities observed ADs, PDs HDs, summarize evidence that clockwork directly influences course disease states. On basis this research, explore circadian-oriented interventions proposed treatments for neurological disorders.

Alzheimer's disease (AD) is a neurodegenerative disorder with growing epidemiological importance characterized by significant burden. Sleep-related pathological symptomatology often accompanies AD. The etiology and pathogenesis of disrupted circadian rhythm AD share common factors, which also opens the perspective viewing them as mutually dependent process. This article focuses on bi-directional relationship between these processes, discussing pathophysiological links clinical aspects. Common mechanisms linking both processes include neuroinflammation, neurodegeneration, desynchronization. Timely recognition sleep-specific symptoms components could lead to an earlier correct diagnosis opportunity offering treatments at stage. Likewise, proper sleep hygiene related ought be one priorities in management patient population affected narrative review brings comprehensive approach clearly demonstrate underlying complexities disruption. Most data are based interventions including melatonin, but larger-scale research still scarce. Following reasoning backed evidence gained from models, novel anti-inflammatory those targeting metabolic alterations might prove useful for normalizing rhythm. By restoring it, benefits would conferred immunological, metabolic, behavioral function individual. On other hand, balanced should provide greater resilience pathogenesis.
