Abstract Background Information The discovery of green fluorescent protein (GFP) and its derivatives has revolutionized cell biology. These proteins (FPs) have enabled the real‐time observation localization dynamics within live cells. Applications FP vary from monitoring gene/protein expression patterns, visualizing protein‐protein interactions, measuring stability, assessing mobility, creating biosensors. utility FPs also extends to biochemical approaches through immunoblotting proteomic analyses, aided by anti‐FP antibodies nanobodies. are notoriously robust with a tightly folded domain that confers strong stability relative resistance degradation denaturation. Methods Results In this study, we report various green, orange, red can be maintained in native, state during entire process sample extraction, incubation buffer, loading, migration on SDS‐PAGE (sodium dodecyl sulfate‐polyacrylamide gel electrophoresis) only minor adaptations traditional protocols. This protocol results ability detect quantify in‐gel fluorescence (IGF) endogenously‐expressed tagged directly after migration, using standard fluorescence‐imaging devices. approach eliminates need for chemiluminescent reagents, as well time‐consuming steps inherent such transfer onto membrane antibody incubations. Conclusions Significance Overall, IGF detection provides clearer data less background interference, sensitivity comparable or better than antibody‐based detection, quantification, broader dynamic range. After imaging, gels still used other applications total staining if needed. It expands possibilities allowing which not available. Our study explores feasibility, limitations, detecting endogenously expressed extracts, providing insights into preparation, imaging conditions, optimizations, potential co‐immunoprecipitation experiments.
