Abstract Background Periodontitis is a chronic inflammatory disease defined by the pathologic loss of periodontal ligament and alveolar bone in relation to aging. Although clinical cohort studies reported that periodontitis significantly elevated males compared females, emerging evidence indicates females with dementia are at greater risk for decreased bone. Objective This study aimed evaluate whether potential sex‐dependent factor using an experimental model induced triple transgenic (3x‐Tg) dementia‐like mice samples collected from senior 65 plus age patients diagnosed dementia. Materials Methods We triple‐transgenic male female age‐matched wild‐type (WT) control ligature placement. Then, osteoclast activity were evaluated micro‐CT situ imaging assays. In addition, we performed dental examinations on Finally, dementia‐associated Aβ42 p‐Tau (T181) osteoclastogenic receptor activator nuclear kappa‐Β ligand (RANKL) gingival crevicular fluid (GCF) measured enzyme‐linked immunosorbent assay (ELISA). Results Alveolar lesions 3x‐Tg but not males, mice. also observed probing pocket depth (PPD) was Using ELISA assay, had levels RANKL GCF samples. Conclusion Altogether, demonstrate have increased males.

Circadian rhythms regulate physiological processes in approximately 24 h cycles, and their disruption is associated with various diseases. Inflammation may perturb circadian rhythms, though these interactions remain unclear. This study examined whether systemic inflammation induced by an intraperitoneal injection of lipopolysaccharide (LPS) could alter central peripheral diurnal neuroimmune dynamics. Mice were randomly assigned to two groups: the saline control group LPS group. The expression clock genes inflammatory cytokines measured hypothalamus, hippocampus, liver. Diurnal dynamic behaviors microglia also assessed. Our results revealed that perturbed gene oscillations Furthermore, trigger neuroinflammation behavior hippocampus. These findings shed light on intricate link between disruption, underscoring significance relation neurodegenerative

Abstract Background Cytokines play an important role in the immunopathogenesis of dental caries. A systematic review and meta-analysis was carried out with following three objectives: 1)To deepen discuss through a comprehensive analysis literature effects caries on activity levels TNF-α, IL-6 IL-8 saliva children young adults, 2)To compare this cytokines exposure group (moderate-severe caries) control (caries-free or mild caries), 3)To determine whether these could be used as complementary clinical diagnostic tool to assess severity Methods The protocol followed PRISMA Cochrane guidelines registered Open Science Framework (OSF): https://doi.org/10.17605/OSF.IO/MF74V . digital search performed PubMed/MEDLINE, Cochrane, Scopus, Google Schoolar databases from February 15th, 2012, January 13th, 2024. methodological validity selected studies assessed using Joanna Briggs Institute (JBI) tool. random-effects model evaluate association between caries/health, concentration IL-8. Results strategy provided total 126 articles, which 15 investigations met inclusion criteria. number patients studied 1,148, 743 represented case/exposure group, 405 group. age ranged 3 25 years. most prevalent cytokine adults active revealed that there are significant differences TNF-α compared their groups. Conclusions findings suggest may have potential biomarkers for assessing severity. However, further research is needed validate larger more diverse populations before application.

Interleukin (IL)-1β is a pro-inflammatory cytokine whose levels are increased in the brains of Alzheimer’s disease (AD) patients. Despite role IL-1β pathology AD, fact that it expressed at very low makes challenging to measure, hence limiting its potential use as reliable biomarker. Moreover, being able accurately and reliably measure IL-1 β blood possible evaluate this biomarker inflammatory response AD. In study, we compared three quantification methodologies, Meso-Scale Discovery (MSD), both V-Plex S-Plex versions, Quanterix’s SIMOA (Single-Molecule Array), serum AD patients age-matched controls. These assays routinely used with high specificity sensitivity human patients, yet best our knowledge, no study has all techniques for their accuracy biomarkers. Our findings indicate two MSD can be control serum, but assay showed highest receiver operating characteristics (ROCs), an area under curve (AUC) 0.9532, which AUC values assay, 0.5660, 0.6632. Taken together, these data show although technologies useful measurement blood, 3.0 more sensitive measuring biomarkers

Alzheimer’s disease (AD) is the most prevalent neurodegenerative disorder, as approximately 55 million people worldwide are affected, with a significant tendency to increase. It reveals three main pathological features: amyloid plaques, neurofibrillary tangles, and neuroinflammation, responsible for changes that slowly lead deterioration of personality cognitive control. Over century after first case report, effective treatments remain elusive, likely due an incomplete understanding precise mechanisms driving its pathogenesis. Recent studies provide growing evidence infectious aetiology AD, hypothesis reinforced by findings beta functions antimicrobial peptide. Among microorganisms already associated Porphyromonas gingivalis (Pg), keystone pathogen periodontitis (PeD), has received particular attention possible aetiological agent AD development. Herein, we review epidemiological genetic linking PeD Pg highlighting identification periodontal bacteria in post mortem analysis patients’ brains identifying putative mechanistic links relevant biological plausibility association. With focus on research shifting from cure prevention, proposed open door unravelling new prophylactic approaches able reduce global burden AD. As hypothesised this review, these could include bionanotechnological approach involving development oral nanoparticulate vaccine based Pg-specific antigens. Such prevent antigens progressing brain triggering pathology, representing promising step toward innovative prevention.
