Critical limb ischemia (CLI) constitutes the most severe form of peripheral arterial disease (PAD), it is characterized by progressive blockade vessels, commonly correlated to atherosclerosis. Currently, revascularization strategies (bypass grafting, angioplasty) remain first option for CLI patients, although less than 45% them are eligible surgical intervention mainly due associated comorbidities. Moreover, patients usually require amputation in short-term. Abstract Background Perianal fistulas (PF) affect one-third patients with Crohn’s disease (CD) limited therapeutic options. There is dearth of literature on safety and efficacy bone marrow-derived mesenchymal stromal cells (BMSCs) in this population. Methods An open-label, phase I/II, single-arm study was conducted involving local administration human allogeneic perianal fistula refractory to standard therapies. Clinical severity biomarkers were assessed at baseline periodically until week 104 , MRI 24 104. Primary secondary objectives assess respectively. Fistula remission complete closure openings < 2 cm collection MRI, response decrease drainage by ≥ 50%. Change activity index, quality-of-life Van Assche index over time using mixed-effect linear regression model. Results Ten (male:8, mean age:27.4 ± 12.0years) recruited. Abstract Background Pulmonary arterial hypertension (PAH) is associated with oxidative stress and affects the survival homing of transplanted mesenchymal stem cells (MSCs) as well cytokine secretion by MSCs, thereby altering their therapeutic potential. In this study, we preconditioned MSCs prostaglandin E1 (PGE1) performed in vitro vivo cell experiments to evaluate effects rats PAH. Methods We studied relationship between PGE1 vascular endothelial growth factor (VEGF) secretion, B-cell lymphoma 2 (Bcl-2) expression, C-X-C chemokine receptor 4 (CXCR4) expression MSC apoptosis migration through hypoxia-inducible (HIF) pathway vitro. The experimental were randomly divided into five groups: (I) control group, (II) monocrotaline (MCT) (III) MCT + non-preconditioned (Non-PC) (IV) PGE1-preconditioned (PGE1-PC) (V) +PGE1+YC-1-PC group. methane dicarboxylic aldehyde (MDA) levels, rat lungs, mean pulmonary artery pressure, systolic right ventricular hypertrophy index, wall thickness index (%WT), relative area (%WA) arterioles. Results Preconditioning increased protein levels HIF-1 alpha (HIF-1α) which can reduce increase CXCR4, migration, secretion.
