Over the past two decades, epigenetic regulation has become a rapidly growing and influential field in biology medicine. One key mechanism involves acetylation deacetylation of lysine residues on histone core proteins other critical that regulate gene expression cellular signaling. Although deacetylases (HDACs) have received significant attention, roles individual HDAC isoforms pathogenesis psychiatric diseases still require further research. This is particularly true with regard to sirtuins, class III HDACs. Polyphenolic compounds have shown promising neuroprotective properties, making them a valuable resource for identifying prospective drug candidates to treat several neurological disorders (NDs).Numerous studies reported that polyphenols can disrupt the nuclear factor kappa B(NF-κB) pathway by inhibiting phosphorylation or ubiquitination of signaling molecules, which further prevents degradation IκB.Additionally, they prevent NF-κB translocation nucleus and pro-inflammatory cytokine production.Polyphenols such as curcumin, resveratrol, pterostilbene had significant inhibitory effects on NF-κB, treating NDs.Recent experimental findings suggest possess wide range pharmacological properties.Notably, much attention has been directed towards their potential therapeutic in NDs Alzheimer's disease (AD), Parkinson's (PD), cerebral ischemia, anxiety, depression, autism, spinal cord injury (SCI).Much preclinical data supporting neurotherapeutic benefits developed.Nevertheless, this study described significance agents, specifically emphasizing impact pathway. This article offers comprehensive analysis involvement NDs, including both clinical perspectives.

Prenatal stress has been reported to harm the physiological and biochemical functions of brain offspring, potentially resulting in anxiety- depression-like behaviors later life. Trans-Resveratrol (RESV) is known for its anti-inflammatory, anxiolytic, antidepressant properties. However, whether administering RESV during pregnancy can counteract induced by maternal unknown. This study aimed assess protective potential against molecular behavioral changes prenatal stress. During pregnancy, dams received 50 mg/kg BW/day orally. They underwent a movement restriction forty-five minutes, three times day, addition being exposed artificial light 24 h before delivery. The male offspring were left undisturbed until early adulthood, at which point they assessments, including open field test, elevated plus maze, forced swim test. Subsequently, euthanized, hippocampus prefrontal cortex extracted RT-qPCR analysis measure Bdnf mRNA expression. By weaning, results showed that led reduced weight gain and, increased these effects attenuated supplementation. findings suggest prevent modulating

Retinitis pigmentosa is a common cause of inherited blindness in adults, which many cases associated with an increase the formation reactive oxygen species (ROS) that induces DNA damage, triggering Poly-ADP-Ribose Polymerase 1 (PARP1) activation and leading to parthanatos-mediated cell death. Previous studies have shown resveratrol (RSV) promising molecule can mitigate PARP1 overactivity, but its low bioavailability limitation for medical use. This study examined impact synthesized new acylated RSV prodrug, piceid octanoate (PIC-OCT), 661W line against H2O2 oxidative stress rd10 mice. PIC-OCT possesses better ADME profile than RSV. In response H2O2, cells pretreated preserved viability more 38% by significantly promoting SIRT1 nuclear translocation, preserving NAD+/NADH ratio, suppressing intracellular ROS formation. These effects result from expressing antioxidant genes, maintaining mitochondrial function, reducing expression, preventing AIF translocation. mice, inhibited PAR-polymer formation, increased reduced TUNEL-positive retinal outer layer, ERGs, enhanced light chamber activity (all p values < 0.05). Our findings corroborate protects photoreceptors modulating SIRT1/PARP1 axis models degeneration.
