Abstract To investigate the effect of Y-27632 on low-temperature metabolism sheep sperm, different concentrations were added to semen at 4 °C in this experiment detect indicators such as sperm motility, plasma membrane, acrosome, antioxidant performance, mitochondrial membrane potential (MMP), and metabolomics. The results showed that addition 20 µM significantly increased integrity rate, acrosome capacity, MMP level, adenosine triphosphate (ATP) total cholesterol content, reduced Ca2+ content. In metabolomics analysis, compared with control group, group screened differential metabolites, mainly involved five metabolic pathways, most significant difference Histidine (P = 0.001). confirmed improved quality preservation under conditions.

Spermatozoa (SPZ) are sensitive to stressful conditions, particularly oxidative stress, which alters their quality; thus, the use of protective molecules as an antioxidant is encouraged. Herein, we used melatonin (MLT) investigate its in vitro effects on human sperm parameters under conditions stress induced by cadmium (Cd). Fifteen semen samples were divided into control, Cd-treated, MLT-treated, and Cd+MLT-treated groups analyzed after 30 min, 6 h, 24 h exposure. Results showed a time-dependent decrease SPZ motility, DNA integrity, increased apoptosis these counteracted MLT co-treatment. Based data, further explored additional just at h. Twenty-one ejaculates (one per individual) were collected split into two aliquots: first used harvest SP assess activity levels OSI, second one cryopreserved. following post-thaw quality parameters evaluated: motility, membrane acrosome integrity, mitochondrial potential, intracellular reactive oxygen species (ROS), lipid disorder. Based on total motility (TM) integrity (SYBR14+/PI−), hierarchically (p < 0.001) clustered groups good (GFE) poor (PFE) freezability. Introduction Ram spermatozoa inevitably produce a large number of reactive oxygen species (ROS) during liquid storage, leading to oxidative stress and decline quality. Therefore, it is particularly important add exogenous antioxidants the process semen preservation. The purpose this study investigate whether adding alpha-lipoic acid (ALA) ram can reduce enhance quality storage at 4°C. Methods Different concentrations ALA (0, 0.025, 0.05, 0.1, 0.5, 1 mM) were added stored During 4°C, motility, kinetic parameters, membrane integrity, acrosome energy metabolism parameters (mitochondrial potential (ΔΨM) adenosine triphosphate (ATP)) [ROS, malondialdehyde (MDA), total antioxidant capacity (TAC), superoxide dismutase (SOD)] assessed. Results discussion results indicated that 0.1 mM significantly ( p <0.05) improved motility (TM) progressive (PM), plasma ΔΨM, ATP, TAC, SOD, while reducing ROS MDA content compared control group. In conclusion, damage caused by in effectively improve preserved And optimal concentration mM.

Reactive oxygen species (ROS) are important factors that lead to a decline in sperm quality during semen preservation. Excessive ROS accumulation disrupts the balance of antioxidant system and causes lipid oxidative damage, destroying its structure function. Curcumin is natural plant extract neutralizes enhances function endogenous enzymes. The effect curcumin on preservation sheep has not been reported. Epicatechin (EPC) is a flavonoid belonging to the family of catechins; it has been described as powerful scavenger wide spectrum reactive oxygen species (ROS) and modulator ex vivo sperm vitality. In this study, we assessed potential protective abilities EPC on cryopreserved bovine spermatozoa. We focused conventional quality parameters, well oxidative profile spermatozoa alongside capacitation patterns, expression profiles proteins involved in process capacitation. Semen samples were presence 25, 50 or 100 μmol/L compared native semen (negative control) ejaculates frozen absence (positive control). A dose-dependent improvement parameters was observed following administration, particularly case motility, membrane, acrosome DNA integrity comparison positive control.
