Randomized controlled trials on diet and shortening of leukocyte telomere length (LTL) mostly focus marine-derived n-3 polyunsaturated fatty acids (PUFA). Walnuts are a sustainable source PUFA. We investigated whether inclusion walnuts (15% energy) in the for 2 years would maintain LTL cognitively healthy elders (63–79 old) compared to control group (habitual diet, abstaining from walnuts). The extraction of bioactive compounds present in walnut (Juglans regia L.) male flowers (WMFs) was performed based on an experimental design using ultrasonic-assisted extraction. Solvent nature, time, and water content were selected as variables, phenolic, flavonoidic, condensed tannins contents antioxidant properties evaluated. Acetone the solvent with highest performance, extracts obtained this displaying increased concentration activities. For several high content, individual polyphenolic tocopherolic evaluated by means LC-MS LC-MS/MS. best conditions for (2.86 mg gallic acid equivalents/g WMF) (29.4 µg/g acetone 40% (N20) 20% (N15), respectively. Although total tocopherol concentrations lower than other Juglans parts, most quantity provided highly biologically active δ-tocopherol (84%). Significant quantities quercetin (101.9 µg/g), hyperoside (2662.9 quercitrin (405.7 isoquercitrin (1293.7 µg/g) determined WMF (N20). Both inhibited enzymatic activity α-glucosidase tyrosinase; however, inhibition observed N20, extract higher content. Conversely, N15 had anticancerous cell lines used, a moderate selectivity towards cancerous phenotype being both extracts. At non-cytotoxic concentrations, displayed good activities cellular cultures, decreasing basal H2O2-induced oxidative stress. Diet is a potential modulator of telomere length (TL), but the impact individual dietary components, such as nuts, on TL in young, healthy individuals remains underexplored. Peanuts are rich bioactive compounds that may influence TL. Therefore, to fill this gap knowledge, study aimed investigate effect peanut consumption specific population. Fifty-eight were randomized one three different intervention groups for 6 months each: (1) 25 g/day skin-roasted peanuts (SRP); (2) 32 butter (PB); (3) control (CB) (based oil). was measured by quantitative real-time PCR saliva at baseline and end intervention. Our findings revealed significant between-group differences changes, particularly between SRP CB over (mean difference: 0.53; 95% CI: 0.01, 1.05; p-value = 0.048).
