The prevalence of inflammatory bowel disease (IBD), which includes ulcerative colitis (UC) and Crohn’s (CD), increases gradually worldwide in the past decades. IBD is generally associated with change immune system gut microbiota, conventional treatments usually result some side effects. Bifidobacterium longum, as colonizing bacteria intestine, has been demonstrated to be capable relieving mice can employed an alternative or auxiliary way for treating IBD. Here, mechanisms longum treatment were summarized based on previous cell animal studies clinical trials testing bacterial therapies. This review will served a basis future research treatment.

Abstract Tumour necrosis factor-α (TNF) is a multifunctional cytokine. First recognized as an endogenous soluble factor that induces of solid tumours, TNF became increasingly important pro-inflammatory cytokine being involved in the immunopathogenesis several autoimmune diseases. In liver, numerous biological responses such hepatocyte apoptosis and necroptosis, liver inflammation regeneration, autoimmunity, but also progression to hepatocellular carcinoma. Considering these multiple functions we propose anti-TNF therapies specifically target signalling at level its specific receptors.

Noise-induced hearing loss (NIHL) is the second most common cause of sensorineural loss, after age-related and affects approximately 5% world’s population. NIHL associated with substantial physical, mental, social, economic impacts at patient societal levels. Stress social isolation in patients’ workplace personal lives contribute to quality-of-life decrements which may often go undetected. The pathophysiology multifactorial complex, encompassing genetic environmental factors occupational contributions. diagnosis screening are conducted by reviewing a patient’s history noise exposure, audiograms, speech-in-noise test results, measurements distortion product otoacoustic emissions auditory brainstem response. Essential aspects decreasing burden prevention early detection, such as implementation educational programs routine primary care specialty clinics. Additionally, current research on pharmacological treatment includes anti-inflammatory, antioxidant, anti-excitatory, anti-apoptotic agents. Although there have been advances understanding NIHL, remain low levels evidence for effective pharmacotherapeutic interventions. Sjogren’s syndrome (SS) is a chronic autoimmune disease accompanied by multiple lesions. The main manifestations include dryness of the mouth and eyes, along with systemic complications (e.g., pulmonary disease, kidney injury, lymphoma). In this review, we highlight that IFNs, Th17 cell-related cytokines (IL-17 IL-23), B (TNF BAFF) are crucial for pathogenesis SS. We also summarize advances in experimental treatment strategies, including targeting Treg/Th17, mesenchymal stem cell treatment, BAFF, inhibiting JAK pathway, et al. Similar to SLE, RA, MS, biotherapeutic strategies SS consist neutralizing antibodies inflammation-related receptor blockers proinflammatory signaling pathways. However, clinical research on therapy comparatively rare. Moreover, differences curative effects immunotherapies among other diseases not fully understood. emphasize targeted drugs, low-side-effect combination therapies should be focus future research.

Tumor necrosis factor-alpha (TNF-α) is a pleiotropic immune cytokine that belongs to the TNF superfamily of receptor ligands. The exists as either transmembrane or soluble molecule, and targets two distinct receptors, TNF-α 1 (TNFR1) 2 (TNFR2), which activate different signaling cascades downstream genes. cellular responses depend on its molecular form, targeted receptor, concentration levels. plays multifaceted role in normal physiology highly relevant human health disease. In central nervous system (CNS), this regulates homeostatic functions, such neurogenesis, myelination, blood-brain barrier permeability synaptic plasticity. However, it can also potentiate neuronal excitotoxicity CNS inflammation. pleiotropism various roles CNS, whether deleterious, only emphasizes functional complexity cytokine. Anti-TNF-α therapy has demonstrated effectiveness treating autoimmune inflammatory diseases emerged significant treatment option for diseases. Nevertheless, crucial recognize effects therapeutic target are diverse complex. Contrary initial expectations, anti-TNF-α been found have detrimental multiple sclerosis. This article focuses describing roles, both physiological pathological, CNS. Additionally, discusses specific disease processes dependent regulated by rationale use target.

Considering the unique therapeutic potential of mesenchymal stem cells (MSCs), including their immunosuppressive and immunomodulatory properties as well ability to improve tissue regeneration, these have attracted attention scientists clinicians for treatment different inflammatory immune system mediated disorders. However, various clinical trials using MSCs purpose are conflicting differ from results promising preclinical studies. This inconsistency is caused by several factors such poor migration homing capacities, low survival rate, level proliferation differentiation, donor-dependent variation cells. Enhancement retention persistent effects remain a challenge overcome in MSC-based therapy. In this review, we summarized approaches enhance outcomes therapy revised current future perspectives creation cellular products with improved diverse applications.

High levels proinflammatory cytokines such as tumor necrosis factor-alpha (TNF-α) interleukins are produced readily by cells to fight Severe Acute Respiratory Syndrome-Coronavirus-2 (SARS-CoV-2) infections.
