Abstract Dry eye disease (DED) affects up to half of the world’s population and causes blur, pain, decreased quality life. Unfortunately, many people do not receive proper treatment due current challenges associated with DED diagnoses. To address this issue, point-of-care (PoC) clinical tear film diagnostics are beginning be used for rapid in-office testing. However, these also have significant obstacles, including sampling. We previously demonstrated how soft contact lenses (SCLs) can capture concentrate proteins on eye. Given low volume subjects, SCL sampling holds great potential improve PoC facilitate prompt patients. Here we show detection new established protein biomarkers using our method. Abstract Dry eye disease (DED) is a challenge in ophthalmology. Rat models represent valuable tools to study the pathophysiology and develop novel treatments. A major DED research detecting multiple biomarkers low tear volume sample. Multiplex immunoassays for rat are missing. We have developed multiplex electrochemiluminescence immunoassay (ECLIA) detect three DED: MMP-9, IL-17 ICAM-1. Tears, used as matrix, were collected from six healthy Wistar rats. Assays run based on U-Plex Meso Scale Diagnostics (MSD) platform, by two independent operators according EMA guideline bioanalytical method validation. Linear mixed, regression fit perform statistical analysis range of concentrations chosen analytes. During optimization, it has observed that incubation time, temperature agitation affected robustness protocol.
