Abstract As the global population ages, bone‐related diseases have increasingly become a major social problem threatening human health. Exosomes, as natural cell products, been used to treat due their superior biocompatibility, biological barrier penetration, and therapeutic effects. Moreover, modified exosomes exhibit strong bone‐targeting capabilities that may improve efficacy avoid systemic side effects, demonstrating promising translational potential. However, review of bone‐targeted is still lacking. Thus, recently developed for applications in this are focused. The biogenesis regulatory functions exosomes, constructive strategies bone‐targeting, effects introduced. By summarizing developments challenges It striven shed light on selection exosome different bone highlight potential future clinical orthopedics.

DNA-based hydrogels stand out for bone regeneration due to their exceptional biocompatibility and programmability. These facilitate the formation of spatial structures through bulk hydrogel fabricating, microsphere formatting, 3D printing. Furthermore, microenvironment can be finely tuned by leveraging degradation products, nanostructure, targeting, delivery capabilities inherent materials. In this review, we underscore advantages hydrogels, detailing composition, gelation techniques, structure optimization. We then delineate three critical elements in promotion using hydrogels: (i) osteogenesis driven phosphate ions, plasmids, oligodeoxynucleotides (ODNs) that enhance mineralization promote gene protein expression; (ii) vascularization facilitated tetrahedral DNA nanostructures (TDNs) aptamers, which boosts expression targeted release; (iii) immunomodulation achieved loaded factors, TDNs, bound ions stimulate macrophage polarization exhibit antibacterial properties. With these properties, used construct organoids, providing an innovative tool disease modeling therapeutic applications tissue engineering. Finally, discuss current challenges future prospects, emphasizing potential impacts regenerative medicine.

Aging is a multifaceted process influenced by hereditary factors, lifestyle, and environmental elements. As time progresses, the human body experiences degenerative changes in major functions. The external internal signs of aging manifest various ways, including skin dryness, wrinkles, musculoskeletal disorders, cardiovascular diseases, diabetes, neurodegenerative cancer. Additionally, cancer, like aging, complex disease that arises from accumulation genetic epigenetic alterations. Circadian clock dysregulation has recently been identified as an important risk factor for cancer development. Natural compounds herbal medicines have gained significant attention their potential preventing age-related diseases inhibiting progression. These demonstrate antioxidant, anti-inflammatory, anti-proliferative, pro-apoptotic, anti-metastatic, anti-angiogenic effects well circadian regulation. This review explores cancers, specific natural targeting key features these conditions.

Melatonin is an important endogenous hormone that modulates homeostasis in the microenvironment. Recent studies have indicated serum melatonin levels are closely associated with occurrence and development of osteoporosis postmenopausal women. Exogenous could also improve bone mass increase skeletal strength. To determine underlying mechanisms prevention treatment osteoporosis, we performed this review to analyze role metabolism according its physiological functions. Serum related mass, measurement which a potential method for diagnosis osteoporosis. has direct effect on remodeling by promoting osteogenesis suppressing osteoclastogenesis. regulates biological rhythm tissue, benefits osteogenic effect. Additionally, participates modulation attenuates damage induced oxidative stress inflammation osteoblasts prevents osteolysis from reactive oxygen species inflammatory factors. As alternative drug can gut ecology, remodel microbiota composition, regulate substance absorption maintain metabolic balance, all beneficial health structure. In conclusion, our systematically demonstrates effects metabolism. Based evidence review, will play more diagnosis,

Abstract Objective To explore the difference in protective effects of intraperitoneal injection exogenous melatonin daytime or nighttime on bone loss ovariectomized (OVX) rats. Methods After bilateral ovariectomy and sham surgery, 40 rats were randomly divided into four groups: operation group (Sham), (OVX), (OVX + DMLT, 9:00, 30 mg/kg/d) NMLT, 22:00, mg/kg/d). 12 weeks treatment, sacrificed. The distal femur, blood femoral marrow cavity contents saved. rest samples tested by Micro-CT, histology, biomechanics molecular biology. Blood was used for metabolism marker measurements. CCK-8, ROS, Cell apoptosis are performed using MC3E3-T1 cells. Results Compared with treatment at night, mass OVX significantly increased after administration. All microscopic parameters trabecular increased, only Tb.Sp decreased. Histologically, microarchitecture DMLT also more dense than LMLT. In biomechanical experiment, femur day able to withstand greater loads deformation. biology experiments, formation-related molecules while resorption-related administration expression MT-1β cell cells treated low-dose MLT had higher viability efficiency inhibiting ROS production high-dose MLT, which turn effectively inhibited apoptosis. Conclusion Daytime acquires better night
