The pandemic situation caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has highlighted the need of fast, simple, and cost-effective tests for diagnosis emerging pathogens. RT-qPCR been established as reference technique SARS-CoV-2 infections. This method requires a time-consuming protocol extraction nucleic acids present in sample. A colorimetric reverse transcription loop-mediated isothermal amplification using calcein molecule combined with simple extraction-free saliva samples (calcein RT-LAMP) developed. Samples are heated 95 °C 10 min before at 63 40 min. results can be observed fluorescence or naked eye color change from orange to green. was compared commercialized available fluorescent RT-LAMP kits. developed shows better sensitivity specificity than commercial same RT-LAMP, without reader. Moreover, has, RT-qPCR, 90% 100% Ct ≤ 34, expensive instruments, demonstrating potential this population screening.
