Immunoassays (IAs) with fluorescence-based detection are already well-established commercialized biosensing methods, such as enzyme-linked immunosorbent assay (ELISA) and lateral flow immunoassay (LFIA). surface-enhanced Raman spectroscopy (SERS) have received significant attention from the research community for at least two decades, but so far they still lack a wide clinical commercial application. This review, unlike any other review that we seen, performs three-dimensional performance comparison of SERS IAs vs. fluorescence IAs. First, compared limit (LOD) key parameter 30 SERS-based immunoassays reported in literature. We also performances smaller number available reports (FIAs). found median geometric average LODs about 1.5–2 orders magnitude lower to immunoassays. For instance, LOD IA is 4.3 × 10−13 M, whereas FIA, it 1.5 10−11 M. However, there no difference relative standard deviation (RSD)—both 5–6%. In this study, we introduce a simplified surface-enhanced Raman scattering (SERS) nanobiosensor for precise detection of SARS-CoV-2 antigen, leveraging supervised machine learning approaches. The biosensor was made with Au nanoislands conjugated 4-aminothiophenol reporter and an anti-SARS-CoV-2 antibody. Through the integration feature selection algorithms, namely, logistic regression, linear discriminant analysis, support vector machine, achieved high accuracies ranging from 96 to 100% in antigen detection. Furthermore, identified underlying mechanisms by employing concept multidimensional calibration space, which is based on decision trees random forest algorithms. This analysis explainable allowed us gain insights into reasons why our exhibits lower sensitivity compared that previous sandwich-type immunosensors SARS-CoV-2. results presented here emphasize potential SERS biosensing, can be applied any type diagnostics.
