Mitochondria are the leading organelle for energy metabolism. The toxic effects of environmental toxicants on mitochondrial morphology, metabolism, and their determination cell fate have already been broadly studied. However, minimal research exists such as pesticides metabolism at in vitro subcellular level, particularly from an omics perspectives (e.g., metabolomics). Here, German cockroach (Blattella germanica) was fed diets with (0.01 0.001 mg/mL) without abamectin, highly purified fat body mitochondria were isolated. Swelling measurement confirmed abnormal swelling caused by abamectin stress. activity two key metabolism-related enzymes, namely succinic dehydrogenase isocitrate dehydrogenase, significantly affected. metabolomic responses isolated to analyzed via untargeted liquid chromatography/mass spectrometry metabolomics technology. Fifty-two differential metabolites (DMs) identified between mg/mL abamectin-fed control groups. Many these DMs enriched pathways involved ATP production consumption oxidative phosphorylation, TCA cycle, pentose phosphate pathway). Nineteen typically related This study is valuable further understanding toxicology under toxicants, its level.

Allium macrostemon Bge. (AMB) and chinense G. Don (ACGD), both named Allii Macrostemonis Bulbus or Xiebai in Chinese, belong to the same genus. They are used as edible vegetables medicinal herbs for treatment of atherosclerosis. So far, comparative analysis spatial metabolomes platelet aggregation function steroidal saponins has rarely been performed. In this study, high mass resolution matrix-assisted laser desorption/ionization time-of-flight imaging spectrometry (MALDI-TOF MSI) were performed on fresh bulbs these two distribution saponins, ultra-high-performance liquid chromatography tandem triple quadrupole (UHPLC/TQD-MS) was quantify their content levels. Platelet inhibitory activity induced by arachidonic acid, adenosine diphosphate collagen fully investigated. A total 53 differential variables with compounds identified tentatively characterized between AMB ACGD samples UHPLC/TQD-MS. Furthermore, almost distributed tunic outer scale regions AMB, while they rich whole leaf scale, developing flower bud bulbs. Quantitative results demonstrated several unique components (macrostemonoside I, timosaponin B-II, macrostemonoside F, etc.) (chinenoside chinenoside II, IV, etc.). Moreover, spirostanol exhibited stronger than furostanol structure relationship further summarized. conclusion, high-mass MALDI full-scan MSI provides abundant information ACGD. And we finished discussion characteristics. These findings would benefit understanding safety effectiveness use.

Understanding the metabolic disorders induced by nano- and microplastics in aquatic organisms at molecular level could help us understand potential toxicity of more thoroughly provide a fundamental scientific basis for regulating usage management plastic products. In this research, effect polypropylene nanoplastics (PP-NPs) (PP-MPs) on metabolites tilapia liver was comprehensively investigated internal extractive electrospray ionization mass spectrometry (iEESI-MS). A partial least-squares discriminant analysis (PLS-DA) one-component variance (ANOVA) were used selecting 46 differential metabolites, including phospholipids, amino acids, peptides, carbohydrates, alkaloids, purines, pyrimidines, nucleosides. Pathway enrichment showed significant effects glycerophospholipid metabolism, arginine proline aminoacyl-tRNA biosynthesis after exposed to PP-N/MPs. Dysregulation these is mainly reflected possible induction hepatitis, oxidative stress, other symptoms. The application iEESI-MS technology without sample pretreatment study under interference provides promising analytical method environmental toxicology research.
