Ferroptosis holds great potential as a therapeutic approach for gastric cancer (GC), prevalent and deadly malignant tumor associated with high rates of incidence mortality. Myricetin, well-known its multifaceted biomedical attributes, particularly anticancer properties, has yet to be thoroughly investigated regarding involvement in ferroptosis. The aim this research was elucidate the impact myricetin on ferroptosis GC progression. present study observed that could trigger cells by enhancing malondialdehyde production Fe2+ accumulation while suppressing glutathione levels. Mechanistically, directly interacted NADPH oxidase 4 (NOX4), influencing stability inhibiting ubiquitin degradation. Moreover, regulated inhibition induced Helicobacter pylori cytotoxin-associated gene A (CagA) through NOX4/NRF2/GPX4 pathway. In vivo experiments demonstrated treatment significantly inhibited growth subcutaneous tumors BALB/c nude mice. It accompanied increased NOX4 expression tissue suppression NRF2/GPX4 antioxidant Therefore, underscores novel inducer interaction NOX4. is promising candidate treatment.

Ferroptosis, an iron-dependent cell death characterized by lethal lipid peroxidation, has been involved in the pathogenesis of various diseases, such as ischemia/reperfusion injury, neurodegenerative and inflammatory disorders. Pharmacological inhibition ferroptosis represents a promising strategy for treatment above diseases. Flavonoids, class natural products found medicinal plants functional foods, are known numerous activities, especially newly discovered anti-ferroptotic activity. Our review aims to expand our understanding flavonoids' activity on multiple biological systems highlight importance utilizing flavonoids ferroptosis-associated A systematic literature search was conducted using Google Scholar, PubMed, Web Science, ScienceDirect, Springer Link. The following keywords: flavonoids, products, ferroptosis, mechanism, available information comprehensively studied compiled. Ferroptosis is an emerging type of regulated cell death usually accompanied by the accumulation ferrous ions (Fe2+) and lipid peroxides. As metabolic hub body, liver crucial for iron storage metabolism. The seems to be closely related ferroptosis through Liver disease greatly threatens host health, exploring effective interventions essential. Mounting studies have demonstrated that one possible pathogenic mechanisms involved in disease. Targeting may provide a promising opportunity treating However, drugs targeting are extremely limited. Therefore, it urgent need develop new safe regulators. Natural active compounds (NAC), especially those derived from traditional Chinese medicine, recently shown great therapeutic potential via modulating ferroptosis-related genes or pathways. Here, we outline molecular mechanism systematically summarize regulatory function NAC on Finally, discuss application prospects problems concerning as regulators managing

Abstract Cancer cells often exhibit defects in the execution of cell death, resulting poor clinical outcomes for patients with many cancer types. Ferroptosis is a newly discovered form programmed death characterized by intracellular iron overload and lipid peroxidation membrane. Increasing evidence suggests that ferroptosis closely associated wide variety physiological pathological processes, particularly cancer. Notably, various bioactive natural products have been shown to induce initiation cells, thereby exerting anticancer effects. In this review, we summarize core regulatory mechanisms multifaceted roles Importantly, focus on regulate such as terpenoids, polyphenols, alkaloids, steroids, quinones, polysaccharides. The efficacy, adverse effects, drug–drug interactions these need be evaluated further high‐quality studies accelerate their application treatment.
