The multivalency of bioligands in living systems brings inspiration for not only the discovery biological mechanisms but also design extracellular matrix (ECM)-mimicking biomaterials. However, designing controllable construction strategies is still challenging. Herein, we synthesized a series well-defined multivalent antimicrobial peptide polymers (mAMPs) by clicking ligand molecules onto prepared reversible addition-fragmentation chain transfer polymerization. multiple cationic ligands mAMPs could enhance local disturbance anionic phospholipid layer bacterial membrane through binding, leading to amplification bactericidal effect. In addition multivalency-enhanced antibacterial activity, enable multivalency-assisted hydrogel fabrication with an ECM-like dynamic structure. resultant self-healing and injectable properties be successfully employed as biomaterial scaffold treat infected skin wounds. strategy presented this work provides new ideas biomimetic highly active biomaterials tissue repair regeneration.

Abstract Irreversible bacterial resistance and impenetrable biofilms have rendered conventional antibiotic therapy ineffective. Nevertheless, increasing evidence has confirmed that carbon nanodots (CDs) exhibit potent antibacterial activity. However, the complex chemical composition of CDs, and, hence, their mechanism, is difficult to understand. The design new CDs against an urgent requirement.In this study, kanamycin‐sulfate‐derived (KCDs) are found significant potential with assistance light irradiation. Systematic analysis indicates KCDs not only retain active structure kanamycin but also produce five components modified on its surface, which shown inhibit activity Staphylococcus aureus beta‐ketoacyl‐acp synthase III (FabH) through molecular docking. Furthermore, multifunctional hydrogels (CG‐KCDs) self‐assemble into injectable, self‐healing, antibiofilm structures noncovalent forces between cationic guar gum (CG). results indicate under laser, CG‐KCDs irradiation exhibits enhanced in vitro by damaging a reactive oxygen species (ROS)‐dependent manner, as determined transcriptomics. Abstract The use of non‐antibiotic strategies to combat refractory drug‐resistant bacterial infections, especially biofilms and accompanying inflammation, has recently aroused widespread interest. Herein, a photo‐therapeutic nanocomposite with bio‐responsive oxygen (O 2 ) self‐supplying is introduced by integrating manganese dioxide (MnO nanozymes onto photosensitizer (indocyanine green, ICG)‐loaded mesoporous polydopamine nanoparticles (MPDA), namely MI‐MPDA NPs. can activate O generation in the infection microenvironment, thereby effectively alleviating biofilm hypoxia. Under near‐infrared light (NIR) irradiation, continuous supplying further boosts level singlet ( 1 ), enabling robust elimination through ‐potentiated photodynamic/photothermal therapy. Interestingly, down‐regulates factor expression inflammatory signaling pathways MnO ‐mediated reactive species scavenging, which ameliorates condition. Meanwhile, prevents M1‐phenotype switch macrophages from overexpression hypoxia‐inducible factor‐1 α (HIF‐1 prompting macrophage reprogramming toward pro‐regenerative M2‐phenotype.
