Colorectal cancer is a prevalent malignant tumor with complex and diverse pathogenesis. In recent years, natural products have shown promising application prospects as sources of anticancer drugs. BBR, class benzoquinoline alkaloids extracted from various plants, widely used in disease treatments owing to its pharmacological activities, including antibacterial, anti-inflammatory, antioxidant, anticancer, anti-angiogenesis properties. Research has demonstrated that BBR exerts an anti-Salmonella -Escherichia coli infection effect, attenuating inflammatory reactions by inhibiting harmful bacteria. During the stage colorectal precancerous lesions, inhibits activity cell cyclin regulating PI3K/AKT, MAPK, Wnt signaling pathways, thereby decelerating cycle progression polyp or adenoma cells. Moreover, inhibitory effect on primarily occurs through regulation cycle, anti-angiogenesis, gut microbiota, antioxidant pathways. The specific involved pathways include MPK/ERK, NF-kB, EGFR encompassing Bcl-2 family proteins, vascular endothelial growth factor, superoxide dismutase. This study reviews summarizes, for first time, mechanisms action carcinogenesis process cancer, providing novel insights clinical intestinal diseases.

Oxidative stress and neuroinflammation are two major causes leading to early brain injury after subarachnoid hemorrhage (SAH). Nuclear factor E2-related 2 (Nrf2) is a critical transcription that contributes antioxidant responses. Additionally, Nrf2 could inhibit transforming growth beta-activated kinase 1 (TAK1), which plays vital role in microglial activation-mediated neuroinflammation. Neferine (NE) exhibits considerable protective effects diverse disease models. However, the detailed effect mechanism of NE on SAH remain unknown. Our data showed treatment significantly reduced behavior cognitive impairment, edema period SAH. In addition, mitigated SAH-induced oxidative damage, neuroinflammation, neural death. Moreover, inhibited M1 polarization enhanced M2 phenotype microglia both vivo vitro. Further investigations revealed Nrf2-antioxidant response element (ARE) signaling pathway suppressed TAK1-NF-κB signaling. contrast, depletion by ML385 Nrf2-ARE signaling, induced activation, further promoted polarization. abated neuroprotective against Notably, LPS also aggravated activation reversed beneficial summary, provides protection inhibiting modulating through suppression.

Ulcerative colitis (UC), a chronic idiopathic inflammatory bowel disease (IBD), presents with limited current drug treatment options. Consequently, the search for safe and effective UC prevention is imperative. Our prior studies have demonstrated that phenolic compound p-Hydroxybenzaldehyde (HD) from Nostoc commune, effectively mitigates intestinal inflammation. However, mechanisms underlying HD's anti-inflammatory effects remain unclear.

Oxyberberine (OBB) is a naturally occurring isoquinoline alkaloid that believed to possess various health-promoting properties, including anti-fungus, hepatoprotection, anti-inflammation, and anti-intestinal mucositis effects. Despite several studies reporting the health benefits of OBB in treating ulcerative colitis (UC), its specific mechanism action has yet be fully elucidated. This investigation designed explore potential protective efficacy latent using an vitro model UC-like inflammatory intestinal cells. Caco-2 cells were pretreated with subsequently exposed lipopolysaccharide (LPS). The transepithelial electrical resistance (TEER), paracellular permeability, distribution expression tight- adherent junction proteins determined assess barrier integrity. levels proinflammatory cytokines, reactive oxygen species (ROS), Nrf2, NF-κB signaling cascade analyzed via ELISA, qRT-PCR, immunofluorescence, or Western blotting. was found mitigate effects LPS on cell monolayers, as evidenced by improvement TEER decrease FITC-dextran flux.
